ABSTRACT

Objective To investigate the effect of lentivirus-mediated shRNA silencing SFRP5 on proliferation,in-vasion and migration of pancreatic cancer cell line PANC-1. Methods A SFRP5-knockdown recombinant plasmid was constructed and successfully transfected it into pancreatic cancer cell line PANC-1,blank plasmid transfection was treated as negative control and untreated cells as blank control group. The expression of SFRP5 at RNA and protein level in cell were detected by real-time PCR and Western blot, CCK-8 assay was applied to examine the effect of SFRP5 silencing on the proliferation, the cell migration of pancreatic cancer cell line PANC-1 was ana-lyzed by Transwell migration assay and cell scratch test was used to examine the cell invasion in PANC-1 cell. Results Stable transfected shRNA-SFRP5 cell of pancreatic cancer line was established successfully. The prolifera-tion capacity of SFRP5 group was significantly higher as compared to the negative control and blank control group by CCK8 assay(P<0.01).Similarly, cell invasion and migration of SFRP5 group were significantly higher compared to the negative control and blank control group(P<0.01). Conclusions SFRP5 lentiviral interference vectors can effectively decrease SFRP5 gene expression in PANC-1 cell of pancreatic cancer, thereby promoting cell proliferation, invasion and migration.

ABSTRACT

Objective To investigate the alterations of the amplitude of low frequency fluctuation (ALFF) by the restingstate fMRI in first-episode major depressive disorder (MDD) patients, and to analyze the relationship between brain area of different ALFF alterations with their aggressive behaviors. Methods Thirty first-episode MDD patients receiving no treatment or stopping treatment for 2 weeks and 30 sex-, age-, and education-matched healthy controls were included in this study. All the participants were scanned with 3. 0 T resting-state fMRI scanner and the blood oxygenation level dependent (BOLD) signals were acquired to calculate the amplitude of low frequency fluctuation (ALFF). Then we examined the differences in ALFF findings between the two groups and the correlations between the scores of aggressive behavior and ALFF in the brain areas showing significant group differences. Results Our results showed that MDD patients had significantly increased ALFF in the left thalamus, precuneus and right lingual gyrus, and fusiform gyrus (P<0.001, corrected by Alphasim). The correlation analysis indicated that the increased ALFF in the left precentral gyrus was positively correlated with the scores of physical aggression, self-aggression, and total score of aggressive behaviors (r=0.41, 0.49, 0.39, P<0.05 or 0.01). The increased ALFF in the right inferior frontal gyrus was negatively correlated with anger (r=-0.44, P<0.05). Conclusion Spontaneous abnormal neural activities exist in some brain regions of MDD patients in the resting-state fMRI, and the increase of ALFF in regions of frontal is closely related to the aggressive behaviors of patients.

ABSTRACT

Sternal malformation/vascular dysplasia association is a rare congenital dysmorphology, which has not yet been reported in Korea. Its typical clinical features include a sternal cleft covered with atrophic skin, a median abdominal raphe extending from the sternal defect to the umbilicus, and cutaneous craniofacial hemangiomata. We report a case of a full-term newborn who presented with no anomalies at birth, except for a skin defect over the sternum and a supraumbilical raphe. Multiple hemangiomas appeared subsequently on her chin and upper chest wall, and respiratory distress due to subglottic hemangioma developed during the first 2 months of life. Her symptoms were controlled with oral prednisolone administration. No respiratory distress have recurred during the 3-year follow-up period.

ABSTRACT

PURPOSE: Magnesium sulfate (MgSO4) is a commonly used drug for eclampsia prophylaxis and the first choice tocolytic agent for preterm labor. Recently it has been reported to have a fetal neuroprotective effect. This study was aimed to evaluate the influence of antenatal magnesium sulfate exposure on perinatal outcomes in very low birth weight infants (VLBWIs) with maternal preeclampsia. METHODS: We conducted a retrospective study of VLBWIs (n=81) with maternal preeclampsia (June 2005 through June 2012), who had been admitted to the NICU at Ewha Womans University Mok Dong Hospital. Clinical characteristics and neonatal complications were analyzed according to the antenatal exposure to MgSO4. Antenatal MgSO4 exposure was the only medication that had been administered up to the period of delivery. RESULTS: Among 81 neonates, 20 neonates were exposed to antenatal MgSO4. Mean gestational age was low in MgSO4 exposed group [29.5+/-2.8 vs. 31.6+/-2.3 weeks (P=0.02)]. The incidence of significant patent ductus arteriosus (S-PDA) was higher in MgSO4 exposed group after adjustment by gestational age (P=0.02). There were no differences between the two groups with regard to rates of respiratory distress syndrome, use of ventilator, retinopathy of prematurity, necrotizing enterocolitis, intraventricular hemorrhage, periventricular leukomalacia and mortality. CONCLUSION: Antenatal MgSO4 exposure was associated with a higher risk of significant PDA in VLBWIs with maternal preeclampsia.

ABSTRACT

Objective To study the relationship between self-esteem and childhood abuse, life events among male violent adolescent. Methods According to the modified overt aggression scale, juvenile delinquents were divided into violent group (n = 128 )and nonviolent group (n = 118 ). All the participants were investigated by Self-Esteem Scale (SES), Childhood Trauma Questionnaire-28 Item Short Form (CTQ-SF) and Adolescent SelfRating Life Events (ASLEC) and conducted correlation and regression analysis. Results ①Violent group had lower self-esteem scores ( 22.73 ± 3.30 ) than the nonviolent group ( 23.81 ± 3.30, P < 0.05 ). Violent group had higher scores in physical abuse, sexual abuse, total life events, interpersonal relationship, punishment, and other (8.30 ±4.07, 7.23 ±2.26, 54.48 ±18.60, 10.09 ±3.84, 14.43 ±5.87, 4.93 ±3.15, 9.93 ±3.64), compared to the nonviolent group (7.27 ± 3.27, 6.60 ± 2.09, 45.40 ± 18.45, 8.42 ± 4.13, 11.07 ± 5.75, 3.66 ±2.81, 7.84 ±3.66, P<0. 05). ②The self-esteem was significantly negative correlated with the childhood abuse and life events( r= -0. 143 ~ -0. 358, P< 0.01 ) among violent male adolescents. ③The physical abuse and punishment had significant prediction function of 15.6% on self-esteem. Conclusions Violent adolescents have low self-esteem, which is closely related to childhood abuse and adolescent life events.

ABSTRACT

PURPOSE: Hand-foot-mouth disease (HFMD) is a common viral illness in children, which is usually mild and self-limiting. However, in recent epidemics of HFMD in Asia, enterovirus 71 (EV71) has been recognized as a causative agent with severe neurological symptoms with or without cardiopulmonary involvement. HFMD was epidemic in Korea in the spring of 2009. Severe cases with complications including death have been reported. The clinical characteristics in children with neurologic manifestations of EV71 were studied in Ewha Womans University Mokdong Hospital. METHODS: Examinations for EV71 were performed from the stools, respiratory secretion or CSF of children who presented neurologic symptoms associated with HFMD by realtime PCR. Clinical and radiologic data of the patients were collected and analyzed. RESULTS: EV71 was isolated from the stool of 16 patients but not from respiratory secretion or CSF. Among the 16 patients, meningitis (n=10) was the most common manifestation, followed by Guillain-Barre syndrome (n=3), meningoencephalitis (n=2), poliomyelitis-like paralytic disease (n=1), and myoclonus (n=1). Gene analysis showed that most of them were caused by EV71 subgenotype C4a, which was prevalent in China in 2008. CONCLUSION: Because EV71 causes severe complications and death in children, a surveillance system to predict upcoming outbreaks should be established and maintained and adequate public health measures are needed to control disease.

ABSTRACT

The management of symptomatic patients with hypertrophic Cardiomyopathy has generally included the use of beta-blockers and calcium channel blockers. Surgical treatment has been attempted for operable patients who have become refractory to medical therapy. However, associated complications, high mortality rate, and high recurrence of symptoms have shifted the treatment focus toward alternative therapy modalities. Recently, a DDD type pacemaker has been introduced as an alternative treatment option. We report a case of a patient with hypertrophic cardiomyopathy treated with dual-chamber pacing.
